Resolution is the distance at which a lens can barely distinguish two separate objects. Resolution is limited by aberrations and by diffraction. Aberrations can be minimized, but diffraction is unavoidable; it is due to the size of the lens compared to the wavelength of the light.

The Rayleigh criterion stated in the equation θ = 1.22 λ/D gives the smallest possible angle θ between point sources, or the best obtainable resolution. Once this angle is found, the distance between stars can be calculated, since we are given how far away they are.

Diffraction limits resolution. For a circular aperture, lens, or mirror, the Rayleigh criterion states that two images are just resolvable when the center of the diffraction pattern of one is directly over the first minimum of the diffraction pattern of the other.

This means that for a standard fluorescence system, such as a confocal microscope, the lateral resolving power is at best limited to around 200 to 250 nm and axial resolution to around 600 nm.

These super-resolution systems have a maximum resolution limit of ~120 nm (as opposed to the diffraction-limited SDCM maximum of ~250 nm). Due to this high resolution, it is necessary to pair these systems with a camera that is also capable of sampling at super-resolution levels.
