The 5′ untranslated region (also known as 5′ UTR, leader sequence, transcript leader, or leader RNA) is the region of a messenger RNA (mRNA) that is directly upstream from the initiation codon.

Creation of a cDNA library starts with mRNA instead of DNA. Messenger RNA carries encoded information from DNA to ribosomes for translation into protein. To create a cDNA library, these mRNA molecules are treated with the enzyme reverse transcriptase, which is used to make a DNA copy of an mRNA (i.e., cDNA).

cDNA (short for copy DNA; also called complementary DNA) is synthetic DNA that has been transcribed from a specific mRNA through a reaction using the enzyme reverse transcriptase. While DNA is composed of both coding and non-coding sequences, cDNA contains only coding sequences.

The resultant mature mRNA, in eukaryotes, has a tripartite structure consisting of a 5' untranslated region (5' UTR), a coding region made up of triplet codons that each encode an amino acid and a 3' untranslated region (3' UTR).

cDNA is derived from mRNA and contain the ORF as well as all or part of the 5' and 3' untranslated regions, to more closely mimic the context of native regulation.

The 5′ untranslated region is the mRNA domain that contains plenty of elements such as upstream ORFs (uORFs), internal ribosome entry sites, microRNA binding sites, and structural components involved in the regulation of mRNA stability, pre-mRNA splicing, and translation initiation.

The synthesized cDNAs are of- ten shorter than the mRNA templates because of various processes that either degrade the mRNA or result in incom- plete transcription. (Note that reverse transcriptase is not made by human cells.

Full‐length cDNAs contain the complete sequence information of their respective mRNA templates. This information is determined by full‐length cDNA sequencing and provides knowledge of the protein encoded by the original mRNA and, in addition, of the untranslated regions (UTR) of this mRNA.
