Nitric oxide (NO) works as a retrograde neurotransmitter in synapses, allows the brain blood flow and also has important roles in intracellular signaling in neurons from the regulation of the neuronal metabolic status to the dendritic spine growth.

Nitric oxide (NO) is an unprecedented neurotransmitter. NO is a small, labile, lipid-permeable free radical molecule. It is synthesized by NO synthase (NOS) and diffuses from its site of synthesis to targets in surrounding cells.

Nitric Oxide (NO) is a particularly interesting example of a neuromodulator because of its very small size and gaseous state.

Its principal receptor is the specialized protein soluble guanylyl cyclase (sGC), which is, in fact, the only known receptor for the NO molecule. Three isoforms of NOS have been identified: inducible (iNOS or NOS-2), endothelial (eNOS or NOS-3), and neuronal (nNOS or NOS-1) — each with separate functions.

In different brain regions, NO and its metabolites have excitatory as well as inhibitory effects. The modulation of autonomic functions by these factors is therefore highly complex and often variable between the different levels from the brain to postganglionic nerve endings.
