Interleukin (IL)-27 is a multifaceted heterodimeric cytokine with pronounced pro- and anti-inflammatory as well as immunoregulatory functions. It consists of the two subunits p28/IL-30 and Epstein Bar virus-induced protein 3 (EBI3).

IL-27, a heterodimeric cytokine of IL-12 family, regulates both innate and adaptive immunity largely via Jak-Stat signaling. IL-27 can induce IFN-γ and inflammatory mediators from T lymphocytes and innate immune cells.

With anti- and pro-inflammatory activity, IL-27 exerts its opposing effects in a cell-dependent and infectious context-specific manner. Upon pathogenic stimuli, IL-27 regulates innate immune cells, such as monocytes, dendritic cells, macrophages and neutrophils.

IL-27 is largely produced by Antigen-presenting cells (APCs) such as Dendritic cells (DCs) and macrophages upon stimulation with TLRs agonists, IFN-α, IFN-γ or microbial infections.

IL-27 suppresses Th2 differentiation by inhibiting GATA-3, and this effect is STAT1 dependent [29]. It also inhibits T cell proliferation and production of Th2 cytokines [34].

The IL-27 signaling pathway can be activated by IL-27 and its receptor IL-27R in different cell types and in different cellular states (Figure 2). In mast cells, IL-27 treatment leads to STAT3 phosphorylation and increases mRNA levels of IL-1α, IL-1β, IL-18, and TNF-α (28).
