Transcription initiation involves the interaction of DNA-dependent RNA polymerase with promoters. In bacteria, this is a highly regulated process. Many regulators interact directly with the bacterial DNA-dependent RNA polymerase, whereas other regulators interact directly with promoters.

Transcription initiation factors (TFII) are proteins which enable RNA polymerase II to bind to the DNA template in order to start the transcription process.TFIIA aids in binding the TATA-binding protein (TBP) to the TATA box containing sequence of the DNA. TFIIB bridges between TFIID and RNA polymerase II.

RNA polymerase and the necessary transcription factors bind to the promoter sequence and initiate transcription. Promoter sequences define the direction of transcription and indicate which DNA strand will be transcribed; this strand is known as the sense strand.

Initiation is the beginning of transcription. It occurs when the enzyme RNA polymerase binds to a region of a gene called the promoter. This signals the DNA to unwind so the enzyme can ''read'' the bases in one of the DNA strands. The enzyme is now ready to make a strand of mRNA with a complementary sequence of bases.

Transcription is the name given to the process in which DNA is copied to make a complementary strand of RNA. RNA then undergoes translation to make proteins. The major steps of transcription are initiation, promoter clearance, elongation, and termination.
